SEM observation for the damage of inner hair cell stereocilia of guinea pig cochlea after loud tone exposure

چکیده
The inner hair cell stereocilia of the guinea pig cochlea was examined under a scanning electron microscope (SEM) after loud tone exposure onto the ear drum of the animal. Before and after guinea pigs were exposed to intensive and continuous tone such as 106 dB SPL in intensity, the functioning of the cochlea was monitored by N1-N2 audiograms. The structural damage of the stereocilia of inner hair cells (IHCs) and outer hair cells (OHCs) was examined using the SEM in x1500 magnification. The preparation of the cochlear specimen is explained in detail. The comparison between the functional change of the cochlea and the structural damage of the IHC stereocilia is done by means of photographic observation. It can be shown that the functional change might be related to the structural damage of the IHC stereocilia after intensive acoustic trauma.

Pure tone overstimulation changes the micromechanical properties of the inner hair cell stereocilia.
The effect of permanent noise-induced hearing loss on the auditory brainstem response (ABR) and the micromechanical properties of cochlear hair cell stereocilia in guinea pigs was investigated. The threshold of movement of the stereocilia was measured by applying force from a fluid filled pipette. After exposure to a 1.0 kHz pure tone signal at 105 dB(A) for 72 h the threshold of the ABR was br...
متن کاملRegeneration of Stereocilia of Hair Cells by Forced Atoh1 Expression in the Adult Mammalian Cochlea
The hallmark of mechanosensory hair cells is the stereocilia, where mechanical stimuli are converted into electrical signals. These delicate stereocilia are susceptible to acoustic trauma and ototoxic drugs. While hair cells in lower vertebrates and the mammalian vestibular system can spontaneously regenerate lost stereocilia, mammalian cochlear hair cells no longer retain this capability. We e...
متن کاملImaging hair cell transduction at the speed of sound: dynamic behavior of mammalian stereocilia.
The cochlea contains two types of sensory cells, the inner and outer hair cells. Sound-evoked deflection of outer hair cell stereocilia leads to fast force production that will enhance auditory sensitivity up to 1,000-fold. In contrast, inner hair cells are thought to have a purely receptive function. Deflection of their stereocilia produces receptor potentials, transmitter release, and action ...
متن کاملA characteristics protein highly expressed in guinea pig inner ear, defined by monoclonal antibody WH-1.
A new monoclonal antibody (termed WH-1; isotype IgG2b) was established using a homogenate of dissected guinea pig cochleas (N = 60) as immunogen. Western blotting and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) identified the WH-1 antigen as a protein or glycoprotein with M(r) approximately 40 kDa. Immunoperoxidase treatment of histologic cryosections of guinea pig coch...
متن کاملThe Effect of Celecoxib, a Cyclooxygenase-2 Inhibitor on Noise- Induced Hearing Loss
Objective(s): Noise-induced hearing loss (NIHL) is the major cause of acquired hearing loss. Celecoxib, a cyclooxygenase-2 (COX-2) inhibitor, is a non- steroidal anti- inflammatory drug (NSAID) with known antioxidant and antineoplastic activity. Therefore, we monitored the extent of temporary noise- induced threshold shifts (TTS) and cochlear damage caused by high level 4- kHz noise exposure t...
